BACs-On-Beads
For detection of gains and losses in selected target regions BoBs™ (BACs-on-Beads) assays offer significant benefits in terms of ease of handling, minute sample volumes, and reduced time-to-result for today's cytogenetics and molecular genetics laboratories. By immobilizing BAC (bacterial artificial chromosome) derived DNA probes onto Luminex® xMAP® fluorescently coded beads, BoBs enable rapid detection of copy number changes in targeted genomic regions from a minute amount of DNA. BACs-on-Beads technology supports high throughput molecular karyotyping in a microplate well which in turn, can lead to greater laboratory efficiency and better use of resources.

      KaryoLite BoBs has been developed to detect aneuploidies, gains and losses in all 24 for chromosomes in a single assay for research use only. The product covers both p and q arms of all chromosomes 1-22, X and Y. It is a bead-based molecular karyotyping assay utilizing probes derived from BAC DNA and coupled to encoded microspheres. It consists of 91 DNA probes immobilized onto polystyrene microspheres distinguishable by the Luminex® instrument system.

      The kit is easy to use with objective and fast result interpretation. The protocol is very robust assuring results that can be relied on. The kit contains sample labeling reagents, the BACs-on-Beads mix, hybridization reagents, streptavidin-phycoerythrin reporter and wash reagents.

      Research may be performed with KaryoLite BoBs using minute amounts of genomic DNA extracted directly from amniotic fluid, chorionic villae or fetal tissue. In fact only 50 ng of DNA is needed, and no cell culturing is required.

      BoBsoft Analysis Software

      BoBsoft™ analysis software provides data analysis for the BACs-on-Beads chemistry products. It takes the multiplexed measurement data in from the Luminex® instrumentation, makes calculations for the user to assess the quality of the assay and calculates the ratios of the patient results compared to normal reference samples. The software presents the calculated information to the user both in numbers and as a graphical plot.

      Major functions of BoBsoft include

      • Subtraction of the background of the negative controls
      • Normalization of the aggregate signal levels between samples and references, and calculation of ratios for all probes for each sample
      • Calculation of overall assay QC metrics from the results of the male and female references

      TriNEST Incubator

      The TriNEST microplate incubator shaker is a high quality device for processing assays on standard-height microplates. Advanced electronic and mechanical design provides the technical excellence you need in order to obtain quality results in either clinical or research assays.

      Compact, versatile solution for plate-based assays, the TriNEST can accommodate three plates simultaneously, and with its footprint requires only 388 x 310 mm of laboratory bench space. You can also stack up to three units to give a nine-plate capacity for the same footprint. The instrument is easy to use, with its own keypad and display. Up to 20 incubating and shaking programs can be stored, in which temperature, shaking speed and time are specified. Interval shaking is also supported. You can specify incubation temperatures from 3°C above ambient to 70°C, a very wide temperature range, making the device suitable for most plate-based assays.

      Controlled incubation and orbital shaking to assure results quality to assure highly repeatable results, temperature is accurate to 0.5°C and temperature uniformity across the plate is good, with variation under 0.6°C. For assays requiring simultaneous shaking, the orbital shaker provides an amplitude range from 400 to 1200rpm, with shaking precision ± 2%.
